Works fine on my test system (Pentium D, fully updated 64-bit SL 4.1):
[root@pr380 ~]# dd if=/dev/zero of=/tmp/pv bs=1M count=0 seek=1024
0+0 records in
0+0 records out
[root@pr380 ~]# losetup /dev/loop0 /tmp/pv
[root@pr380 ~]# pvcreate -v /dev/loop0
Set up physical volume for "/dev/loop0" with 2096768 available sectors
Zeroing start of device /dev/loop0
Physical volume "/dev/loop0" successfully created
Any "avc: denied" messages in your logs?
